Formulation and in vitro analysis of gastro retentive drug delivery system containing timolol maleate

The objective of current research work was to formulate sustained release matrix tablet of timolol maleate using natural and semi-synthetic drug release retardant polymers. Guar gum, xanthan gum and hydroxy-propyl methyl cellulose (HPMC) were used in different concentration in formulation of timolol maleate. Nine formulations were formulated by using direct compression method. Gastroretentive floating tablet of timolol maleate employs polymers (guar gum, xanthan gum, HPMC) and gas generating agents (sodium bicarbonate, citric acid). FTIR showed no drug polymer interaction. All formulations were evaluated for pre-compression (bulk density, tapped density, Hausnerís ratio, compressibility index and angle of repose) and post-compression parameters (hardness, thickness, diameter, swelling index, weight variation, friability, disintegration and dissolution). All results were significant and within range. Among all formulations, F2 containing 37.5% guar gum, float in 20 s showed 71.33% drug release up to 8 h. Most of formulations (F1, F2, F4, F8 and F9) showed zero order drug release. Formulations F3, F5 and F6 showed first order drug release.
